CALCIUM EFFLUX AND SECRETION OF a-AMYLASE FROM RAT PANCREAS
نویسنده
چکیده
1 The efflux of 45Ca from rat pancreas was investigated in relation to the secretion of c-amylase. 2 The 45Ca space in the pancreas was increased 1.6 times when pancreatic tissue was incubated in media in which the extracellular calcium concentration was reduced from 1.0 to 0.05 mM. 3 Carbachol increased the rate of 45Ca efflux from the tissue and this effect was associated with an increase in the release of a-amylase. Dibutyryl cyclic adenosine monophosphate (AMP) also evoked a secretory response, but did not alter the rate of 45Ca efflux. In combination with carbachol, the dibutyryl analogue of cyclic AMP reduced the carbachol-stimulated increase in 45Ca efflux while enhancing the release of secretory protein. 4 The stimulatory effect of carbachol on 45Ca efflux was observed when pancreatic tissues were incubated in media containing a high concentration of ethyleneglycolbis (,B-aminoethyl)N,NA-tetraacetic acid (EGTA).

Calcium efflux was estimated in is...
متن کاملStimulus-secretion coupling in the developing exocrine pancreas: secretory responsiveness to cholecystokinin
We have studied the onset of secretory responsiveness to cholecystokinin (CCK) during development of the rat exocrine pancreas. Although acinar cells of the fetal pancreas (1 d before birth) are filled with zymogen granules containing the secretory protein, alpha-amylase, the rate of amylase secretion from pancreatic lobules incubated in vitro was not increased in response to CCK. In contrast, ...

متن کاملCalcium metabolism and amylase release in rat parotid acinar cells.
1. A method is described for the isolation of rat parotid acinar cells by controlled digestion of the gland with trypsin followed by collagenase. As judged by Trypan Blue exclusion, electron microscopy, water, electrolyte and ATP concentrations and release of amylase and lactate dehydrogenase, the cells are morphologically and functionally intact. 2. A method was developed for perifusion of aci...
متن کاملCalcium fluxes in isolated acinar cells from rat parotid. Effect of adrenergic and cholinergic stimulation.
Rat parotid acinar cells dispersed by a combination of enzymatic treatments remain sensitive to adrenergic and cholinergic agonists. Previous studies have implicated Ca2+ in both adrenergic and cholinergic responses. This paper describes the effects of adrenergic and cholinergic stimulation upon 45Ca2+ fluxes in isolated parotid acinar cells. Suspensions of dispersed cells took up 45Ca2+ from t...
